Structural malformations of the heart are the commonest abnormalities found at the time of birth and the incidence is higher in fetuses that are lost during the first trimester. Although the form of the heart has been studied for centuries, it is in the past decades that the genetic pathways that control heart development have been unraveled. Recently, the concept of the second heart field, a population of multipotent cardiac cells that augment the initial simple heart tube, has clarified the development of the heart. Understanding how the second heart field is used in morphogenesis and how genes interact in a subtle and more complex way is moving us closer to understanding how the normal heart forms and why abnormalities occur. In this chapter, we present a description of the morphological processes that create the formed postnatal human heart and emphasize key genetic pathways and genes that control these aspects. Where possible, these are also linked to the common patterns of human cardiac malformation. Undoubtedly, the details will refine or change with further research but emphasis has been placed on areas of greatest certainty and the presentation designed to promote a general understanding.

Neuroferritinopathy remains the only autosomal dominant syndrome of neurodegeneration with brain iron accumulation (NBIA). While the majority of identified cases appear to be part of an extended pedigree in the northeast of England, patients are increasingly being identified across the globe. Since its discovery in 2001, there have been significant developments in our understanding of the pathological, radiological, and clinical aspects of the condition, though several key pathomechanistic questions, and crucially treatment paradigms, remain unaddressed. This chapter summarizes the genetic etiology, pathological, radiological, and clinical data from all published data to date and suggested potential new avenues for therapy.

Small non-protein-coding RNAs play an instrumental role in both regulating gene expression (creative influence) as well as suppressing selfish genetic elements (defensive role). There are three main groups of small RNAs including microRNAs, endogenous siRNAs and piRNAs. MicroRNAs represent the most important gene regulatory small RNAs and act by tuning the expression level of eukaryotic mRNAs. Interestingly, endogenous siRNAs as well as piRNAs apparently serve both regulatory and defensive purposes. They suppress the expression of repetitive DNA elements but also influence the expression of protein coding genes. For endo-siRNAs, intriguing roles in epigenetic regulation are emerging. The multiple tasks of small RNAs are in line with a role as drivers of organismal complexity.

In addition, they can be used to study gene duplicationand/or deletion. Although they are used in studies on microorganisms includingfungi, bacteria, protists, and archaea, it appears that these genetic markers are not beingutilized to their full microbiological potential. Microsatellites have many advantages over other genetic markers currently in use as they are in general species specific, andtherefore, cross-contamination by nontarget organisms is rare. Furthermore, microsatellitesare suitable for use with fast and cheap DNA extraction methods, with ancientDNA or DNA from hair and fecal samples used in noninvasive sampling, making themwidely available as a genetic marker. Microsatellites have already proven to be a usefultool for evolutionary studies of pathogenic microorganisms such as Candida albicansand Helicobacter pylori, and the onset of new sequencing techniques (such as 454,PACBIO, and mini-ion sequencing) means the ability to detect such markers willbecome less time consuming and cheaper, thus further expanding their potential toanswer important microbial ecology questions.

The cardioprotectivc actions of the fish oil (FO)-derived long-chain n-3 polyunsaturated fatty acids (LC n-3 PUFAs) eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) have been demonstrated, and dose-response relationships have been defined. However, there is a substantial and well-recognized within-population heterogeneity in response to FO, the etiology of which is poorly understood. Genetic variation may influence responsiveness. Here we review the available literature relating to gene variants shown to influence tissue LC n-3 PUFA status and response to FO intervention. From this review we conclude that the available evidence is relatively limited. A number of individual genotype x LC-n3 PUFA x phenotype associations have been described, but few have been investigated in subsequent cohorts or confirmed in independent studies. in the context of a more stratified approach to the provision of dietary advice, there is a need for further research to refine current dietary EPA and DHA recommendations.
